Experimental testing of 199Tl-diethyldithiocarbamate radiopharmaceutical for assessing brain perfusion

Purpose of the research are to study pharmacokinetics of 199Tl-diethyldithiocarbamate (199Tl-DDC) for evaluating the possibility of using in for brain perfusion scintigraphy, to assess its toxicity, and estimate radiation doses for critical organs. Experiments were carried out with 85 albino rats of both sexes, 5 dogs, and 8 male Chinchilla rabbits. Blood and visceral radiometry was carried out by Tracor Analytic gamma-spectrometer 1, 5, 10, 20, 40, 60, and 120 min after intravenous injection of 199Tl-DDC in a dose of 5 mBq/kg. Dynamic and static scintigraphy of the viscera was carried out using Omega-500 gamma-chamber. 199Tl-DDC starts accumulating in the brain immediately after injection and its level remains rather high over 2 h. Radiation dose for critical organs are 4-16 times lower than those after injection of 201Tl-labeled analogs. Toxicity studies showed no notable changes in visceral structure and functions after a single intravenous injection of 10-fold diagnostic dose of 199Tl-DDC. The agent is recommended for clinical trials.
